What is PT-141 (Bremelanotide)?
PT-141, also known as Bremelanotide, is a synthetic peptide and an analogue of the α-MSH hormone (α-melanocyte-stimulating hormone). It is being studied in scientific research, particularly for its ability to activate melanocortin receptors in the central nervous system.
The action of PT-141 is primarily linked to the MC3R and MC4R receptors, which play a central role in the regulation of sexual motivation and libido. Initially developed as a derivative of Melanotan II, clinical research has revealed that it has specific effects on sexual function without influencing skin pigmentation.
Potential Benefits
PT-141 can increase sexual desire in men and women by activating melanocortin receptors in the central nervous system. Studies show improved erection and better sexual response in men suffering from erectile dysfunction. Unlike PDE-5 inhibitors, PT-141 acts centrally in the brain and not through a direct influence on blood flow, which makes it potentially effective even for sexual dysfunctions of psychological origin. The effect can manifest 30 to 60 minutes after application. PT-141 is also being studied in the context of hypoactive sexual desire disorder (HSDD), particularly in post-menopausal women.
Mechanism of Action
PT-141 works by activating MC3 and MC4 melanocortin receptors in the hypothalamus. This central action distinguishes PT-141 from many other substances that primarily act via peripheral vasodilatory mechanisms. Activation of the MC4 receptor can not only increase sexual motivation but also have positive effects on emotional state and mood.
Dosage and Usage
Typical starting doses range from 400 µg to 700 µg. The effect can last 1 to 6 hours, or even up to 30 hours depending on the protocol. A common regimen: 8 units up to 7 hours before sexual activity, preferably in the evening, with an optional additional 5 units in the morning. One pen contains 200 units of 50 µg each. Examples: 1 unit = 50 µg — 5 units = 250 µg — 10 units = 500 µg — 12 units = 600 µg.
Common Combinations
PT-141 is often combined with Melanotan II in the context of sexual health and libido, and with Ipamorelin to analyze energy, regeneration, and physical performance.
Possible Side Effects
PT-141 shows good tolerability in studies. The most common side effects are: headaches, nausea, increased blood pressure, and transient facial flushing. These symptoms are generally mild and transient.
